Both ? and * mean one wildcard tile. LETTER? has seven tiles, including two Es and two Ts; the blank can supply one extra letter, not an entire word.
The default uses every tile. Include shorter words to use a subset, then refine with length, prefix or suffix filters. Matches use a general English dictionary, not a licensed game lexicon.

Yes. Enter ? or * for one blank tile, up to two blanks. Each blank supplies one letter. Repeated letters need repeated tiles or blanks: LETTER contains two Es and two Ts.
No. Starts with, Ends with and Must include only restrict the results. Those letters must already be available in your tiles or supplied by blanks. Filters accept letters A–Z; put blank symbols in Your letters.
Each game chooses its own accepted-word list. These helpers use a general English dictionary that includes uncommon words, so a match is not a guarantee that your game accepts it.
